I think you hit the nail on the head with the Oculus Rift support comment.
There have been enough rumours that Sony is introducing their own 3D head mounted display. I would not surprised if Sony introduces it as a PS4 accessory at E3. I don't see Microsoft developing their own headset, so a partnership with Oculus Rift makes sense.
Aside from that, if this is supposed to be a huge surprise, would be a FF7 remake timed exclusive. Square-Enix could use the money, and Microsoft could use the hype. If it happens, I don't think it would sell well (<5 million on XBOne), but the hype would give a nice boost in sales, especially for a Japanese launch.
MS throws a metric ass-ton of cash at Nintendo to bring some Nintendo games to the X1, agrees to help Nintendo on future handheld/tablet platform. That would be the "biggest surprise ever" and turn the console war on its head, but these things rarely live up to the hype.
Microsoft buys the copyright to Cranky Kong, incoming XBox One exclusive spinoff title.
But seriously, MS's main advantage is that it is the only parent company of the 3 that have dozens of billions to spare and is by far the most stable. Pulling in 5 billion NET every 3 months.....where does it go?
If MS wants to get serious with the Xbox division, and their acquisitions/hiring/buyouts tell me they are, then they should just go all in and buy up a major player in gaming. Literally force the naysayers to either buy your console to play their favorite IPs, or tuff luck. Gamer boycotters never stick to their word....for more then a few days lol.
If KOTOR3 happens, I will literally buy a One after the show.
let's be honest,... nothing will ever be as surprising has MS confirming xbone's rumored DRM policies last E3.
So this would have to be bigger than the GTA4 multiplat announcement, or the Final Fantasy XIII multiplat announcement.
There aint gonna be anything bigger. Not unless they straight up went out to buy a big 3rd party developer.
