to get wolf (if you've beat SSE, use your beaten save file) go back to the ruins stage and there will be a door at the bottom of a scrolling elevator section, go in there and fight wolf and beat him.

Jiggly puff, go to the swamp level (after beating SSE) and go to the swamp level. there will be a door floating in mid air that has jiggly in it. Play for a win to get jiggly.

Toon link will be in the forest stage.

I got Jigglypuff after clearing Event #25 (I think 25 was the number).

You have to either beat SSE or play a lot of VS matches to unlock Jigglypuff, Toon Link, and Wolf. If you beat event 20 after finishing SSE then Jigglypuff will unlock. Before then she won't for whatever reasons.

It's sad.  In the past two games, I always seemed to unlock the Jiggle first but Brawl has changed that... now Jigglypuff is unlocked last (for me).

I have a question. Does online matches help to unlock characters or only offline matches?

 Ok thanks. I guess I'll have to unlock all the characters by the SSE because I already have over 300 matches, but 200 of them are online.
