Well, Nintendo winning E3 2006 was part of the reason they're now the market leader in current gen consoles with the rep they were able to build up. Remember how Sony considerably fucked up, well that's whats helped put them in the position they're slowly rebuilding from.
Simply put, Nintendo NEED to 'win' E3 this year to keep the momentum going with the media and with that the consumers, I think they know this and I think they're gonna pull out some huge stops to really impress! If they do that and put a good show on like they did back when Miyamoto came on stage and composed to Wii Music (come on, you know they won it from that moment on!), then they're set for another year!
Sony and MS will be the heated battle. I don't know why, and I have no justification for this, but I just feel that MS might be having some big weapons up their sleeve, maybe software or hardware wise?
