Skyward Sword and the Last Guardian are up there as well, though the complete and utter silence surrounding both titles has kept my excitement somewhat low so far.

Pretty easy question for me.  Definitely Legend of Zelda: Skyward Sword.

Deus Ex 3. And not even close. Wait, Arkham City. Okay that's close.

So many games coming out. Batman AA, inFamous 2, LBP 2, Deus Ex, Uncharted 3, Portal 2,etc. It's too hard too pick.  

Do I have to play Mass Effect 1 to enjoy Mass Effect 2 and 3? I'd like to get into the series but ME1 isn't releasing on PS3 and I doubt my Labtop is powerful enough to run it. :(

EDIT: Screw it, I'll get it anyway, Bioware said that they would inform us on the important stuff,

1: Skyrim

2: Uncharted 3

3: Gears 3

It's a close call between Uncharted 3 and Mass Effect 3, but I'm going to have to go with Mass Effect 3.

Diablo III, easily.

Followed by Skyrim, Uncharted 3, Forza 4 and Heroes of Might & Magic 6.

Mass Effect 3, nothing else is even close. Gears 3, Dead Space 2, Dragon Age 2, Dota 2, Skyrim follow afterwards in no particular order.
