Ok, this year at E3, we all believe that the first party linup from Nintendo will contain the most surprises, that's given.
Studios like Retro, and N-Keyoto have projects they are working on, obviously, and Reggie has said that this year we will have a "new Nintendo game" before Christmas.
So, its safe to say we'll get at least one new Nintendo game this year.(Lets hope that its not the Mario Baseball game but lets also hope that the Mario Baseball game comes out this year.)
Besides Nintendo's big surprise 1st party announcments, we also have several 3rd parties announcing things on the Wii at E3.
For sure we have Capcom

EA who has said they have new 8 games in the works for the Wii, yesterday on Kotaku(look it up yaself!)
and
Square-Enix who is obviously working on a few new unannounced projects for the Wii, according to their words and the obvious inferment of the White Engines port to the system.
Then we have companies like Activision and Ubisoft who have pledged support to the Wii already, but will in all likelyhood cement that support at E3, given their games are far enough along.
Next, we have Factor 5 who is working on a new "secret" game for the Wii and we have no idea what it is.
Sega will, in all likelyhood debut a new Wii game, though this is speculation.
But I have some more broad assumptions here:
Big first party games from Sony and MS will amount to things we already highly suspect:
God of War 3, almost certainly from Sony.
Now, MS could surprise the show with Halo 4 footage, but this is highly unlikely, and its possible we won't see any big games from their first party showing this year.
3rd party support will be as wide ranging as always, but I expect the brunt of the impact will come from the appearance of the Wii as a much more considerable player amongst 3rd parties this year at E3. It will certainly be the Buzz on the showfloor, as this is the first year the Wii has "proven" itself with consistant hardware and software sales.
Therefore, by deduction, I believe that the majority of "new" announcments for games on consoles will be for the Wii, DS, and PSP, while the majority of big 3rd party games for other consoles will have been seen previously to the show(with several exceptions, of course).
Certainly we know the "big" games for 360 and PS3 that will be prominant at the show, from lots of developers, like Final Fantasy XIII will likely be prominantly on display in SE's PS3 section.
Therefore, I think the majority of "exciting" or should I say "newsworthy" announcments will include titles yet unannounced for the Wii console. While there is no sure way to predict this, and it is almost certain all consoles will have plenty of major announcments, currently, I can safely predict that if anything is gonna beat the Wii in "major announcments" from a software perspective, its gonna have to have one hell of an E3.
