Nope the multiplayer is local only but that's not what Punch-Out!! is about.

I'll be playing tonight JSKI probably, that's if your willing to play with a scrub like myself without no contesting if you think my win was undeserved.

It's really up to you, I didn't know punch out was this close to release, thought it was after E3; now I get why people are so interesting in E3.
