In a nutshell, the Nintendo Network will basically replace the Nintendo Wifi connection and will offer online play, dlc and entire downloadable games like XBL and PSN. It will be supported by the 3DS and Wii U and will release in parallel with Square Enix's Theatrhythm which will offer the first DLC content through the nintendo network.

Wow finally, thank you Nintendo, let us all rejoice :)
