hasn't happened to me lately
This happened to me a few times early on. It was especially frustrating when one of the posts was comparable to a page in Word.
Well, I just did this about five minutes ago (was editing a post in the "Super-Official-ous Wii games of 2010", so it was a biggie...) and I accidentally missed a click only to see the work literally disappear. I was pretty peeved and took a 5 minute break before starting again. 
This wasn't my first time, and it probably wont be my last...

I was upset at first, because I wrote a fairly long follow-up post. Then I made the miraculous discovery that I could open another Reply window, hit Ctrl Z(Undo) and most of my post reappeared! Give it a try(with something simple) and see if it works for you.
Possibly relevant info: Windows 7 64-bit and Google Chrome.
EDIT: Confirm it works for me with Firefox as well. Let's spread the word, don't ever let a misclick equal a non-post again.
Yes, it has happened to me...
It seems to me that the number of forum posts is significantly down, and this is probably one of the causes.
EDIT - @Linkzmax: Indeed that seems to work here too, Ubuntu 64 bit and Firefox.
