I'm guessing the big focus will be on Star Fox Zero. It seems to be a potentially important game for reviving the series and tending to the fans for a while. Besides that, I'm thinking a bunch of release dates and trailers of smaller games. Bravely Second, Hyrule Wariors Legends, Detective Pikachu, and so on.
They could also use the Direct to announce their Zelda anniversary plans. It would be just in time for Twilight Princess HD, and we'd probably get Zelda U info in that case. If we get a game announcement, I guess Paper Mario 5 would be a fairly safe bet. It may be for the 3DS again though.
