All most anything you want, since it is your preferance. Textures, Environment, Quests, Weapons, Clothes, in game characters, Music, physics, menus, almost everything. Even vanilla is quite versatile; I for one deplore the hand to hand combat so I typically go the mage/archer/assassin route. You can poison people by pickpocketting them, or control them by enraging them and making them attack their allies.

Its very versatile if you immerse your self and allow the game to take you places instead of the main branch quest.

One mod I really like is alternate start: http://www.nexusmods.com/skyrim/mods/9557/? allows you to start the game in a variety of different situations.

My 5th character was a merchant who lost everything shipwrecked in the sea above winterhold. In combination with my other mod, Frostfall, which has the ability of making frigid water lethal, I had to get out of the shipwrek through the sea and onto land without sucumbing to frostbite, something extremely hard shipwrecked in a sea in winter. Her aspiration was to become the emperor of Skyrim, (another mod), which required 50,000 gold and an assassination of the leaders of the warring sides of Skyrim.

My 6th character was a new recruit for the Dark Brotherhood (assassin's guild), due to another mod Installed earlier I was able to become a custom race called mindreaver which has the ablity to either calm, control enemies, and make them run away, but only some enemies of certain kinds, so this new character is built around sneaking and backstabbing people, but if I'm not careful I could attack the wrong person and end up getting killed cause I can't fight head on. To make things even more intresting, to fast travel in the game you have to visit a place at least once. Since I started the game as the dark brotherhood, I had nothing on my map so I had to trek across Skyrim to reach remote destinations and assassinate people, even going so far as to plan routes so that I end up back at HQ. While treking I assassinated random people, both good and bad, cause I'm basically RPing as my character who became an assassin because of her insatiable drive to kill.

Not to mention there are many full voiced custom follower mods, with alot of personality like Vijla, Sofia, Nadina, Sara, Inigo and endless other ones such as those included in Intresting NPCs mod.

In fact imo, the main branch quest is the worst and the most boring.

Currently, I have 181 mods

35: armor/clothing (uptextured/redesigned vanilla armors, fancy clothes [my current assassin character wheres a kimono], any accessorie you can think of [I had a DBZ scouter once]

13: body mods (custom hair, custom bodies, tattoos, remaster textures)

18: Combat (changes to combat including various new monsters and mob AIs, I have one that controls the civil war and bandit spawns)

38: NPCs (additional intresting NPCs, custom followers etc)

17: quests

4: Race mods

25: system mods (one changes my load screens to feature watercoller paintings of skyrim, my main menu and menu theme are completely different, Sky UI completely overhauls the inventory and magic menus, mod manager manages mods.)

51: Unassigned (it takes time to put them in categories)

18: World mods(like modifiying the moon the environment, like tropical skyrim (I prefer my skyrim cold thoguh), changes to weather lighting etc.

Skyrim with Mods is a whole nother game entirley, a much better one IMO.
