I would have to go with Okami.
Yeah, yeah, I know... It's a port of a PS2 game. But that PS2 game was an amazing game, and with added Wii features such as true Widescreen and Wiimote functionality, the Wii-make turned out to be a masterpiece. The learning curve can be pretty tough, but once you get past it, battling foes and using the Celestial Brush is a flawless experience.
The gameplay is tried and true, but at the same time goes into new territory for a simply refreshing experience, the graphics are simply gorgeous, the bosses are larger than life (Nechku & Lechku are probably my favorite bosses this generation. I mean, steampunk demon owls? Yes please.), and the story has many a touching moment, whether you're laughing or getting teary eyed.
Okami is simply a game that cannot be missed, whether you own a PS2 or a Wii.
