First of all, it's absolutely unrealistic to think a game like metroid could sell 20m like Mario Kart or Animal Crossing.
Adventure sci-fi just doesn't have that kind of universal appeal across all demographics.
The best realistic expectations imo would be to achieve Zelda (pre-botw) kind of reach. So in the order of 5 to 8 million.
The main problem is Metroid's main peculiarities (level design, game progression, atmosphere) are hard to market for people who have never played the series so far.
The best think you could do to reach a wider audience imo is mixing core Metorid design with something people are already more used to / concepts they understand better. A natural choice - considering the nature of the game - would be giving it a more survival and sci-fi horror theme. Both concepts are already very popular in gaming and in media in general.
If you manage to market the game as a sci-fi horror survival/adventure while retaining all the key level design and progression qualities of classic Metroid you could really reach a wider audience imo.
