I think they will do the same thing they did to the Wii:
Double the processing power,
Double the Graphics power,
Add some motion control (tilt control),
Improve the voice rec.,
Add a SD slot for DLC (no HDD),
Improve the touch screen(s),
Improve the Wii comm. for DLC and DL games,
Make it DS compatible,
Add a internet browser,
Make the screens bigger (but the DS2 will be smaller),
And add some Nintendo Magic ?????.
