Here's my take on a realistic roster: 40 characters
Take the Brawl cast (35) and cut:

What do you mean by Wii Sports integration? Like giving a Mii a Fighting Sword (from Wii Sports Resort) or a Bow and Arrow(Also from WSR)?

Sorry, but I'm going to choose 5 "characters" that aren't from Nintendo, and a total of 35 (plus an extra 5 if we allow another 5 non-Nintendo characters, for fun). I don't see why this isn't plausible, especially since Brawl had 37 characters. Anyway, my list will include a variety of "alternate forms". These are alternate versions, where the moveset is mostly the same, but with power/speed/jump/etc modifications. Not all characters will have alternate forms, only those that have obvious alternates. Multi-character sets (eg Ice Climbers, Pokemon Trainer) are considered to be one character.
The hero: Mario (w/ Dr Mario alternate form), Link (w/ WW Link alternate form), Samus (w/ Zero Suit alternate form), Kirby, Fox (w/ Falco alternate form)
The antagonists: Bowser (possible Baby Bowser alternate form), Ganondorf, Wario (possibly Waluigi alternate form), King Dedede, Metaknight
The support crew: Peach (daisy as alternate costume only), Luigi, Zelda/Sheik (w/ Zelda/Tetra alternate form), Yoshi, Diddy+Dixie Kong
The extra protagonists 1: Ice Climbers, Pit, Marth (w/ Roy alternate form), Ness (w/ Lucas alternate form), Olimar
The extra protagonists 2: Mr Game and Watch, Captain Falcon, Donkey Kong, Shulk (possibly with Ike alternate form), Mii (w/ Animal Crossing protagonist alternate form)
The pokemon others: Pokemon Trainer, Jigglypuff, Pikachu (w/ Pichu alternate form), Lucario (w/ Mewtwo alternate form), Blaziken Krystal
The visitors: Sonic + Tails, Snake, Mickey Mouse, Rayman, Mega Man
The extra visitors: Alex Kidd, Ryu Hayabusa, Ryu (w/ Ken alternate form), Sora, Rabbids
Since there were 35 choices in Brawl, 35 is the number I went with here, too (the extra visitors aren't included - they're just the "if they're going to add a few extra...). Note that the alternate forms would be selected like costumes, and would be near-identical in terms of animations, etc (except perhaps for the entry and taunt movements).
EDIT: You'll notice I left off ROB. I don't think he really fits in. For the same reason, I left off Bayonetta - I don't think she'd really fit into the game well.
EDIT 2: Damn, forgot Krystal. Ah well, drop Blaziken in favour of her.

Thought it would be better to keep the lists shorter. But if there's 35 in Brawl it would make sense to change my OP!

I am not one for adding in more 3rd party software characters from diffrent series. I beleive it will ruin the feeling of the game taking away to much attention foucsed on a nintendo fighter game.
and the next smash needs more vilians.
