Tingle's Rosy Rupeeland. Seriously, a lot of people make fun of it, but it's honestly one of the freshest, funniest, and most original games I've played in recent years. Lasts a long time, too.

Some other great games:

Castlevania - Order of Ecclesia - just has that old school vibe to it, and one of the few very challenging games I've played this gen. Good amount of content, even after you beat the game.
Rythm Heaven - Really fun and unique rythm game. Kinda like warioware mixed with guitar hero (the drum part)
Big Brain Academy - surprisingly fun, also very challenging, at least to earn the platnuims. Really works the brain..
Scribblenauts - Has a few flaws and akward controls at first, but I love the concept of solving puzzles by conjuring up virtually any item you want.
Animal Crossing - Really fits with the DS style of games
Clubhouse Games - lots of solid mini games
Meteos - If you like puzzle games check this one out, makes good use of the touch screen.

I hear the Dragon Quest remakes are good stuff as well if you like RPGs, but I haven't played them yet.

Have you tried the Hard Mode Lvl.1 cap? It's pretty much one of the hardest challenges I've ever had in my entire video-gaming life. And I've went through the classics, Ghouls and Goblins, Battletoads, Contra, you name it. 
If you want a bigger challenge though, try doing that mode, and get the boss medals at the same time XD It will drive you insane, especially the last bosses. 

Oh man, I did the hard mode for a bit, just on the first level I started to get my ass kicked. I even played as Albus, who is a beast, but I still died often. It took awhile to get used to him. I think I beat a few stages in that, but eventually just stopped playing out of frusteration and moved on to new games.

Albus is indeed a beast, especially using his shot Torpor combo. Got through most bosses with it xD 

But yeah, Hard mode is too frustrating, especially on the boss battles. Blackmore alone took me more than 2 hours of trial and error to defeat him.

Lunar Knights-Non-canon sequel to the Boktai series, you play as dark swordsman and a solar gunslinger and switch off on the fly to hunt down vampires.  Lots of fun and you can do a lot of weapon upgrading/side questing.

Lost Magic-Draw runes on the screen to do spells, combining different runes creates new spells.  A really cool combat system.

Beware of this game... I like it... but at least to me, not good at music, it's impossible! I mean it, impossible.

Too bad, because the game is very fun.

If you like strategy/puzzle games then get Scribblenauts. It's awesome. One of my favorites. If you don't know what it is, here's a summary: You are Maxwell, a character with a notepad that you can use to spawn anything you want in order to solve a level. Like seriously, anything. If you want a chupacabra, just type it in and there it is. Magic Carpet? Check. Griffin? Check. Mattress? Check. Any noun you can think of, you can write it down in the game and it will appear. There are over 22,000 words recognized by the game. It is truly amazing.  In order to solve a level, you must obtain a Starite (a star-shaped object). There is a Puzzle mode and an Action mode. Puzzle levels involve problem solving, such as returning a tiger cub to its mother while avoiding a dinosaur. But that's easy stuff. This game can get really challenging. Anyway, the Action levels simply involve the Starite sitting somewhere in the level, and you have to use your imagination in order to get it while dealing with the obstacles in the way. For example, if a dragon is blocking your path to the Starite, you can spawn a Shrink Ray, shoot it at the dragon, and it will shrink! Then you can go merrily to your Starite. There are about 220 levels, and there are plenty of hidden achievements to unlock in the game. I'm a little over halfway through the game, and I'm having a blast. If you like Scribblenauts, then look out for Super Scribblenauts ( the sequel) this fall, which has tons of new features.
