Realistically, you'll need multiple playthroughs just to find the harder routes. And if you've never played the game before there's a high chance you're going to get a game over on your first run. Star Fox is a bit like a "rogue-like lite": There's many different routes you can take. For example you could go easy-easy-easy in one playthrough and then easy-medium-easy in the next and medium-hard-medium on the third.
Also, your three companions can go down in every mission (and will go down on your first couple of playthroughs) thus missing the rest of the stage and not being present in the stage after. Most likely you'll lose Falco in stage 1 on your first playthrough and he'll only be back in stage 3, etc. So there's dozens and dozens of variations of how a run can play out.
Also, it's an arcade game. It was made to be replayed and replaying levels is a lot of fun because you get better each time and uncover new stuff in every stage. Playing the game gets more fun the more you play it.
