Trigun.

If you haven't seen the series, you should, but I think it's well structured to be made into a very nice game with only the basic amount of conversion required. Of course, depending on how close it followed the story, it might require some serious on-rails gameplay.

It's got an excellent mix of seriousness and silliness, but it also has a fairly decent set of pre-established "boss" battles for the game to be built around. In some ways it almost comes across as a more innocent version of No More Heroes, and a similar graphical style could work well for it.

Aside from Vash, you also have Wolfwood(he doesn't show up until episode 9, but you could still show his story before that point) and then possibly Millie and Meryle that could each have their own viewpoints throughout the story, sorta like how in sonic adventure(the original dreamcast one) you could play through as each new character you bumped into. At the same time though, it wouldn't be required to play through as all  of them if you didn't want to.

About the only real problem is getting the player to accept an "absolutely avoid killing, but be ready for it" mentality. With most games, the entire point is to kill the badguys, and that's just what you do, otherwise, if you aren't supposed to kill people in the game, you're never even allowed to at all. With Trigun, the focus would be to NOT kill people, despite the fact that you do still need to shoot at people and at one point, if following the anime, kill. In order for that to work, you'd need to be able to kill, but somehow make it so the goal was still to avoid killing. It's possible this could be done through limiting the ammo available.

I see it utilzing 3rd person shooter controls similar to what I've seen in Tomb Raider: Legend, but done with Wiimote&Nunchuck, or maybe even a never before done Wiimote& Classic Controller setup (either classic controller held in both hands, with Wiimote attached to aim, or classic controller in one hand and wiimote in other hand)

The graphics wouldn't have to be that intense to get the feel of the anime, and the anime already has a lot of repeating character models and very barren landscape. It should be well within the capabilities of the Wii, and if it was too big to fit the whole series on a single disc(No More Heroes was awfully short), I think it would be well worth either having a second disc, or doing it in two parts.(1st set of 13 episode on part 1, 2nd set on part 2)
