Nintendo needs to re-establish F-Zero as a popular franchise again. After that, they can try and make it an E-sport, but I don't see it happening.
Nintendo has two of the biggest E-sports around already: Pokemon and Smash. And Splatoon has the potential to evolve into something way deeper and competitive. They have that front covered.
