A few months ago, I might have said RPG. But Monolith seems to have that covered pretty nicely. Monolith has the ambition and the talent to make great software. Action-adventure is well covered. Zelda and Metroid are two of the best. Platforming is beyond covered. Strategy, simulation, fighting, and racing are doing OK. So that leaves the elephant in the room, the one genre Nintendo is most sorely lacking and the one least likely to be made because of its design philosophy: shooter. If Retro is indeed working on Star Fox, they would have a new rail shooter, but they'd still be missing a third or first-person shooter. Something like Jet Force Gemini or Perfect Dark, to name two N64 gems. I've written this several times here: Nintendo should go after ex-Eurocom employees and create a new first-party studio dedicated to a new shooting franchise. It doesn't need to be as violent or controversial as Gears of War or Call of Duty -- since this is something that clearly weighs on the minds of Nintendo executives -- but it would fill a rather large gap in Nintendo's portfolio.

Yeah, shooter was my first thought too. But the market is already over saturated with shooters, and sony and microsoft already have their shooters, so why would people move over to nintendo? That shooter would in that case need to be a serious halo killer.
But neither sony or microsoft have a massive, open world western rpg. And they have an visual impact that few can represent. So that is my bet.

The shooter market isn't saturates on nintendo that is the point. There are people who still own one console each generation.

Good point, nintendo needs a shooter to, I just think that a western rpg should come first. But that thought is a bit biased since I don't really like shooters :p
