If it's a straight up 2d platformer like New Super Mario Bros was (you can't see the ground as it's perfectly flat), then trying to add some kind of depth to that would probably get confusing and make the game overly chaotic.

If it's 2.5D (slight perspective on the ground), like for example the 2d segments in SMG2 or Donkey Kong Country Returns, then I could see the player being able to navigate up and down almost like they would in a classic arcade brawler.

Still, I personally would prefer it to be a straight up 2d platfomer with any 3d effects not tampering with the perfection of 2d Mario's gameplay.
