Non-spoiler version: about 2/3 of the way through the main quest
Spoiler version: After your father dies and you enter the Brotherhood of Steel.
In either case, head back to Megaton at the right time, and you'll receive a radio broadcast. Listen to it in its entirety to trigger the mission.
