In the non canon movie, he's in his 40s, and the creator said he's in his 20s. I wonder who we should trust more.

The mustache is something that can be grown with enough dedication. Trust me, I've seen mustaches like that.

He's not really saying Mario is that age tbh only that the age was the only thing at the time that they considered would be in his design. It's very likely Mario doesn't have a concrete age now that his basic design is settled on.
