Mario Galaxy isthe best game i have ever played....so it's my choice.....also Metroid Prime 3 and Call of Duty 4

I have to break mine up also, single-player and multi-player are different experiences.

Single: Portal (just edging out Mass Effect)
Multi: Rock Band

Super Mario Galaxy
Runner Up: Orange Box (Just for Portal)

Phoenix Wright 3 was prob the most fun I had with anything. Then SMG. Hotel Dusk was godlike too -- the industry REALLY need more well-written, stylish and creative titles like this. Along those lines, at least people paid attention to BioShock.

Okami is a great game but that is the same with SMG.
