I'm going out on a limb here and saying that not only can Nintendo win E3, but they will.
I think the strongest Press Conference will Not be Nintendo however. Nintendo Focuses on the media outlets that are not game specific during the conference, CNN, FoxNews, AP, Reuters. With that said, they will absolutely win the large and vast casual audience that got to such places for entertainment news. MS vied very strongly for this crowd today and failed miserably.
Casual Winner = Nintendo
During the Conference; Nintendo will be doing something that they have done before. They will blow out software. They are being criticized for their perceived software deficiencies and to combat it they will highlight tons of software. Nintendo did this back in the mid-NGC years with fairly good results. This year they will have better support from 3rd Parties, especially Japanese, and many 1st party surprises. Literally we could have a year in which we are shown Kid Icarus, a new Retro tile, Animal Crossing, New Zelda, Pikman, Final Fantasy 7 remake, KH 3, Two Tales exclusives, Atlus Exclusives, Nipon Ich projects, Level 5 projects, EA projects, Activision Projects, Disaster, NST project, Namco projects, and Independent ventures like deBlob and Conduit. That really does trump MS's heres four sequels and some crappy casual rip-offs with one surprise announcement. In Sony's defense, they will also be blowing out games like Nintendo, but I believe Nintendo will walk away with more interesting and better titles.
Software Winner = Nintendo
As for Hardware..... w/e I'm not getting into this
