I know it's kind of cheating... But I think it's one of these three then:
Wolfenstein 3D, Duke Nukem 3D or Doom.
I hope it's the first one, because I think that's the best one of those three, but I don't think so since that one didn't redefine it, but rather made the FPS genre. So... I'm gonna say, Doom.

Nope, Korppi got it. It's the original Half-Life.
Before Half-Life all FPS copied the formula of the 3 games you mentioned above. Then Half-Life came along and suddenly characters, believable AI and storybuilding all became staples of SP FPS campaigns.
