Last gen almost all big franchises such as Mario and Zelda demostrated why the Wii mote was essencial for the gameplay of their titles.
For instance, Mario Galaxy implemented the use of the Wii mote briliantly to the game mechanics from collecting coins to swinging Mario. How can nintendo EAD take full advantage of this gamepad?
However, unlike the Wii where the Wii mote made sense and the 3D on the 3DS adds depth to the gameplay. I don't see the point of the WiI U gamepad, it just seems like an add on peripheral.

They could have used it to manipulate the level. Implement Pushmo/Pullblox-like puzzles, tap the screen to reshape the level geometry around the point you tapped etc.
MANY possibilities.

yes they could,but they won't .
As soon as they do so people will cry out for more manipulation.Something like lbp.Than nintendo must release their level editor to the common people.Than many levels will ne made by gamers and interest in mario games will drop because of fatigue.Instead of 10 units they may only sell 1mio.
