5 million seems high. I think 2 million is a minimum, just given all the promotion I've seen Nintendo doing for this title.
I have no idea how popular Punch-Out!! has ever been in Japan, but 3-3.5 million lifetime seems achievable, although there are a awful lot of good games coming in the next few months which could sap Little Mac's endurance.
