Most of the Lucasart's stuff, Monkey Island, Day of the Tentacle, etc. Those were genuinely amusing games.

Space Quest 6 had some LOL moments for me. Sam and Max (old and new are pretty good). Cat Mario (http://files.filefront.com/catmariozip/;9346957;/fileinfo.html) is quite funny ... in a strange, sadistic way. Portal had its moments.

Out of all of 'em? I'd say that Conker's Bad Fur Day is definitely the most hilarious I've played so far. Although titles like Mario & Luigi as well as No More Heroes have some great dialog. Oh, and can't forget TimeSplitters... "It's time to SPLIT!"
