The first XBO update you had to manually turn on your XBO if it was off, so if you usualyl turn it on through voice it wont work that way.  Im not sure about if your XBO is on already though. I assume it would be simlar to how windows works and that you have to wait until you restart your computer for it to happen.  Though i doubt itll give you a notification.  It may say something like you cant play on XBL until updating or something like that though.
