As soon as I read the title, I was thinking about Africa and, since I wrote a paper on it, Leopold II and the Belgian Congo.
As you say, binary, the treasure angle is obvious. The idea of tying in the source of the Nile hadn't occurred to me - excellent idea.
Only difficulty I can see is that it's quite recent and therefore enjoys less mystery.
Perhaps go back several centuries earlier and follow Vasco da Gama?

Some great ideas here, and they make me want Uncharted 3 all the more. I look forward to seeing which historical figure/quest ND settle on and setting of on another great adventure.
I hope they employ even more use of flashbacks/cinematics as I thought that worked very well. It would also allow, if they wanted, to show more of Nate and Sully's past via flashback levels so long as they related somehow to the current plot.
Try to be reasonable... its easier than you think...
