All the bosses and most common foes in MDK were freaks. The obese giant final boss even had a dispenser full of his favourite snack, some sort of one-eyed alien baby-seals. 
In comparison Duke Nukem 3D is probably a lot more amusing overall, but few foes are freaks, many are true monsters and the humour mainly comes from Duke's attitude, towards them, himself and in general.
