Personally, while I don't like the idea of Nuzlockes, I did try doing a Nuzlocke Challenge twice - both times in Black 2, and both times ending in miserable failure after a series of critical hits at the first gym. (Tackle+STAB+Crit at the beginning of the game is brutal)

That said, with the much needed nerf of critical hits in the 6th generation, I guess I might think about this. Still, I'm a bit (very) unsure about the idea of doing a Nuzlocke on my first playthrough of a game.

And what's wrong with Nuzlockes? Is it the unfair chance of criticals or difficult gym leaders, or is the idea of it too cruel to do in a game like pokemon.
