Would it make sense for Microsoft to make Minecraft 2 Xbox exclusive?

It should launch on Xbox first, other consoles later, and they should charge Sony/Nintendo an extra royalty. In this case, the more people play the game, the better. Minecraft 2 won't do nearly as well as the first so they should get it into as many hands as possible.
