You can bet your bottom dollar that Sony will have options for 1 or 2 handed play. They took the same stance with Move games on PS3. So I would say you will be able to play with one hand and the DS4 can replace the move.

Think about the practicality of playing that with the Dualshock, regardless of whether they allow it or not.
