The joke isn't that Mr. Smithers is gay.
The joke is that Mr. Burns can't tell.
But either way, he might not actually be gay. He could be straight but with an insane lust for one man that he can't understand or control, kinda like how I feel about David Bowie or Shigeru Miyamoto. Maybe that's why he's so depressed?
