"Let us respect people's opinions, and just because someone says no, let us not assume they are transphobic or phobic-anything.
For example, there are some roles which favour females (e.g. receptionists) and it doesn't mean the manager is misandrist, just that the manager wants a female facing their clientel."

Um, yeah it does mean that the manager is transphobic. Unlike working at Hooters or something, there is no job that "favors" non-trans people. That doesn't exist. It's explicitly and unapologetically transphobic.

Unless that "job" is in a strip club, Hooters, or any other organization that profits specifically from exploiting the sexuality of their consumers and could argue that they'd directly lose business from their customers not being sexually attracted to a transexual, it's textbook transphobia.

And for the record, if any man with the right credentials thought he was denied a receptionist job purely based of him not being a woman, even when he was perfectly qualified, he most definitely would have a case to sue the company for misandry.
