It kills me having to narrow this down to one game. I'll go with an X-Box classic that is backwards compatible with the 360:
Guilty Gear XX #Reload=It's STILL the definitive version of Guilty Gear as it's the only version with online multiplayer, regardless of the fact that it's been redone multiple times since then. The game has quite possibly the greatest art direction in 2D i've ever seen, has a vast amount of original characters that ooze personality and is the competitor's fighting game with a huge learning curve.
