In my opinion, it won't be a mainstream game that we're all expecting. Perhaps a new series will pop up under our noses and surprise us. Don't just guess random games that are selling well.
You make a good point about Brawl. I thought Rock Band got robbed from GOTY contendership by many publications just because it was a rhythm game. Considering that by this gen's end we'll probably look back and see Guitar Hero and Rock Band being what defined this generation it's funny that neither has ever gotten any GOTY awards.

 


How can Guitar Hero define this generation if it's been coming out for PS2 since before the 360 came out?

 

On topic, it sounds like MGS4 to me as of right now.

It doesn't matter when a series was created when deciding if it defined a generation. GTA III partly defined last generation but has it's roots in the PSOne. A few issues ago in Game Informer they had a small article showing each generation and what defined them for consoles. Here's what they said:

Arcade Era: Space Invaders and Pac Man
8-bit: Super Mario Bros and the Legend of Zelda
16-bit: Super Mario World, Sonic the Hedgehog and EA Sports
32/64-bit: No clear winner. They describe it as a fallen king with many successors fighting over the crown. They name games like Gran Turismo, Resident Evil 2, Metal Gear Solid, GoldenEye 007, and Ocarina of Time vying for the crown with no clear cut winners.
128-bit: Halo, Grand Theft Auto and Madden

Now this gen I think would be defined by the war between Rock Band and Guitar Hero.

 


Yeah, it's not like the greatest game of all time didn't come out in that era, Ocarina of Time (even if you don't like the game you have to respect it).  Definately no winner there.
