Could it be the première trailer for Zelda U? Sure. But I doubt it will be.

Why? Because Miyamoto has always been the one to show off new Zelda games. If they were planning on showing Zelda U to people, they'd have Miyamoto there, not Reggie.

X is a bigger possibility, in my opinion, but I don't think it'll be that, either. A real possibility is Bayonetta 2, although I'm not sure what the Video Game awards could do for Bayonetta 2 that E3 2013 didn't do.

For some reason, my instinct is telling me that it'll be a new IP, being worked on by Nintendo and some third party, perhaps even a third party that nobody expected to be working on a Wii U game (like, for instance, Valve, or Epic Games). Why? Because otherwise, Nintendo would probably just show it off via a Nintendo Direct. This has to be something that they need to show to gamers who aren't paying attention to the Wii U. Which also, in my opinion, rules out DKC, Yarn Yoshi, and other such games.

The people who watch the Video Game Awards are almost all the kinds of gamers into FPS, dark and gritty games, highly-realistic graphics, etc. And it needs to be an announcement that might make the viewers specifically want to buy a Wii U, so it needs to be either completely exclusive or with exclusive features so good that anybody who can afford a new gaming console would buy a Wii U for it.

At least, that's my opinion. I guess we'll see in a few days.
