Looking at her picture on twitter, I would be hard pressed to figure out her gender upon a glance. Could be someone who identifies herself as a female, could be someone who is a male crossdresser (not quite the same thing) and so on.
Clearly though, she is not a biological female, and a lot of people don't have experience dealing with that. I wasn't there and don't know what happened, but if I was to take a guess, the host probably just didn't know how to handle the situation and made a poor choice of words.
If you're a transgender person, that's good for you, but you have to understand that it's something unusual to a lot of people. You shouldn't tolerate disrespect, but you have to be understanding of people who are unprepared and act in a well intentioned manner that makes you feel uncomfortable. Just my two cents.
