Super Metroid is great for speed-running. In fact most Metroid games work well.
Sonic the Hedgehog is made for speed-running. Just keep running!
2D Mario also works very well. It really shows in many areas that the designers considered speed-runners when they made the levels.
Lastly, Donkey Kong Country (Returns) is another must-have for speed-runners. Hard as hell but very satisfactory.
