In terms of hours played probably Monster Hunter Tri and Team Fortress 2.
But my favorites were Mass Effect 2, Mario Galaxy and Zelda Twilight Princess (I know the game was on the GC but I played it on the Wii). Runner ups are: FE: Radiant Dawn, Skyward Sword, Rayman Origins, Galaxy 2, VVVVVV and Dishonored.
