Nintendo can't win the next E3 unless both Msoft and Sony screw the presentation of their new consoles (like Nintendo did last year, but worse).

What they can do, however, is show everybody that they are not only relevant but also a competitor to be feared. And the only way to do that is

1-Getting every multi-platform third party game that gets announced or shown in more detail (like Star Wars or WatchDogs) during the show on WiiU.

2-Show in some form or another their big cards.

The first point is the harder one but also the one that could benefit more Nintendo as that will reasure many WiiU owners and may give an important push to those on the verge of buying it.

The second one is not only easy to achieve but there have been hints of Nintendo trying to achieve it. For instance, from some comments it's pretty clear that the new Smash will be there, maybe only in the form of a trailer but there is the chance of having a small demo. Then it's also likely that Zelda WiiU appears somehow (like the artwork that lead to Skyward Sword). And then there is Monolith, Retro and Platinum Games with Bayonetta 2.

If Nintendo can achieve these two points, then it won't matter what the competition do or who wins the E3, Nintendo will be ina very good situation.
