With each entry in a fighting game franchise new characters are added, this is why SC IV have more characters than SC II. The extra characters that SC IV have didn't exist when SC II was made.

A more interesting question would be why Soul Calibur IV is missing several modes that Soul Calibur II have like Time Attack, Survival, Team Battle and Team Battle VS.

 

Now onto why Assassin, Berseker, Lizardman, Sophitia and Seung Mina would be DLC if SC II were made today.

SC II appeared first in the arcades without those characters. It got patched four times (versions A, B, C, D) before the console release.

Sophitia and Seung Mina won a popularity contest in Japan and were added as secret characters to the japanese version of the game.

Assassin, Berseker and Lizardman were added to the game in the american and european versions.

 

If SC II were made today we would get the first and buggy Soul Calibur II version A without those characters. If it sold enough then it would get patched and Sophitia, Seung Mina, Assasssin, Berseker and Lizardman would be added as DLC.

Tekken 6 existed only as an Arcade game long before it came to consoles and even then they released it with the Bloodline Rebellion update included on PS3 and Xbox360.

But Namco doesn't give the same treatment to Soul Calibur and Tekken. Soul Calibur doesn't get arcade releases anymore.
