Significant to the storyline? No not at all. It does provide story for the characters of Ezio and Altair though. The main story of the AC universe is pretty much entirely ignored. In fact a pretty hefty chunk of the game is either tutorial missions or only related to goings on in that city.
It's an okay game but BY FAR the weakest out of the console versions. I would say AC2 >> AC:B >>>>> AC >>>> AC:R
AC:R has the worst story by far, some of the worst gameplay elements (Desmond, Den Defense etc etc) and depressingly the absolute worst city. There's no variety at all. No countryside, barely any different areas of the city, no horses etc etc. It seems like since AC2 the series has been going backwards in this regard.
The good things is it's still the great core gameplay and some of the missions themselves are very well done.
