I would like it to be in further future like 50 years after Mass Effect 3, so you could have some of the old Crewmembers like Liara, Wrex and Grunt but still have a fresh start. And it would be cool to see some influence from your earlier decisions like the Krogan or Quarian/Geth decision. The most obvious things are a great gameplay and Story but Bioware has proven that they can handle that. I just hope it won´t be a MMORPG. And alltough I was dissapointed by the ending of ME3 i would preorder ME4 immediatly.

There won't be a post-ME3, especially since I don't think they'll change the "destroy all remnants of Reaper technology" scenario, which to be quite frank, should be the only scenario available for any kind of conclusive ending of the ME series. Every species of the galaxy must find their own ways and carve their own paths both in technology and evolution, and they can't do that as long as they depend on the technology left by the Reapers, or nothing would change at all. 

So, imagine a post-ME3 galaxy, with no immediate point to point travel like the Relays, and it would take centuries via FTL flight to get from one system to the other. Somehow that won't do too good of a ME game, me thinks
