It's hard to explain exactly what you do in the game as there is so much to do and many different ways to play it. Basically you collect different blocks by mining, choping, digging etc. and then you can use those blocks to build pretty much whatever you want. During night monsters spawn so you have to build a shelter or a fortress to protect yourself.
The official trailer gives a general idea.
If you want to try a very basic version of Minecraft you can play Minecraft Classic for free. Bear in mind that the full version has a lot more content.
