You don't have to, but only a few hours into the game it will without a doubt make ME2 better.  I've only just got past the Archangel part and if I hadn't played the first game that part wouldn't be nearly as awesome if I hadn't played the first game. 

As a bonus, you also get some bonuses at the start of the game if you import a saved game from Mass Effect.

Mass Effect is pretty cheap whether you buy it for the 360 or PC and if you don't side quests and play it on easy you can breeze through it pretty quickly.  I think my playthrough this weekend took about 15 hours but I also did all the Citedel sidequests and some others.

So yeah, you can play Mass Effect 2 without playing the first, but that would be a giant disservice to yourself.  That would be like watching The Two Towers without watching The Fellowship of the Ring.
