I think it's up to us to find our own meaning in life. I don't think anyone can give a real meaning that applies to everyone. (Ignoring the biological point of view, that is. 
)
As for an after life, I am a Christian so I do believe in life after death. Again, this is a highly subjective matter, so it's really up to you to decide what you believe.
