I have so many favorites... but picking quickly on the humorous side I've always enjoyed Alex Baldwin's line from Working Girl:
'Who the fuck died and made you Grace Kelly?'
I love that it doesn't make sense explicitly, yet you know exactly what he means.

I don't watch much TV, but I'll back up SciFiBoy with one of may favourite Star Trek lines from the 'Devil in the Dark' episode:
'Dammit, Jim, I'm a doctor, not a bricklayer!' - at least I remember him saying bricklayer or builder or something similar.
No wait! Futurama! Pretty much every line in the 'Amazon Women in the Mood' episode... particularly:
'...you're killing me... (violent blow with a club to the head follows statement) Arghh. You're really killing me!'
Try to be reasonable... its easier than you think...
